When this system works properly, the immune response is targeted, proportionate, and self-limiting.
In lupus, multiple layers of immune regulation fail simultaneously. The body does not clear apoptotic (dying) cells efficiently, leaving cellular debris β including DNA and nuclear proteins β in circulation. The immune system produces auto-antibodies against these nuclear components, creating complexes called anti-nuclear antibodies (ANA) and anti-double-stranded DNA (anti-dsDNA) antibodies. These immune complexes deposit in tissues throughout the body β in the kidneys, blood vessels, skin, joints, and brain β triggering complement activation and destructive inflammation.
